About

PCR reaction is used in molecular biology  to make  copies  of  DNA-molecules, encoding our  genes.   To run  the classic version of such reaction  one  need a PCR-machine,  essentially  thermocycler.

Existing DIY versions  are too complex  to make  by  amateur  biologists.

So, I have made  a special  design  of PCR mashine  that is easy  to reproduce  everywhere  in the world  with minimal knowledge  and  skills in electronics. No Arduino , no thermostat modules, pure  steam-punk  version.. made from recycled parts. And  very  easy in case of  repair  and maintenance.

It is  functional  ,  and educational  as well.

It can be made  by school-children  in science-class   from the  materials  found in   trash-can  in the nearest FabLab,  and   Law of Om  and  a little bit of Thermodynamics..

It can be run  full-manually , or  using a 3D printer as automatic  hand.

It involves  the participants  into the magic of science.

The result of that project  is going to be  a video  with demonstration  and instructions  how  to build up  your own  thing  like that  and how to use it ...   And  text  also  of course...It is Open Project... everyone  can make  own version  based  on the  principles and using  things  that are  readily  available.

I am always  open  for  personal communication about  it.

Speaking about scientific research, that system in some situations could provide huge advantage over standard robotic PCR-thermocycler with one block of metall changing temperature by the program. It is just proof of principle , universal machine.. It can be extended to 12 chambers or more, just need a bit of experimentation with resistors to tune them for different T values .. and all the temperatures are available at the same time. I would love to use such full-manual PCR-machine at my work as R&D researcher in lab. I always tried to do something strange and new there. Unfortunately I did made it only now, when I have lots of free time and no job to invest my creative energy into. I see some interesting possibilities when that system might be superior to standard thermocycler.. For example micro-volumes PCR, or when very fast transfer between different temperatures needed. For robotic commercial thermocycler it is next to impossible to move temperature from 99C to 1C in a few seconds.... With this machine it can be done easy.. And so on. This universal device has no limits at all, everything is possible. Some extra manipulations during PCR are also possible, like microwave or light treatment of samples, or just add one simple fluorescence reader and it becomes Real-Time PCR... It has potential not only for DIY amateurs but also for professional labs too, when prototyping new methods, or a protocol for example

Risks and challenges

The prototype exists, just need some extensive testing and compare with professional machines and summary of it. I need to write a G-Code programm for 3D printer to move head in between 3 positions.. and so on ... lots of small things... But principal is done. I can manually stabilize the temperature of vessel with oil quite good. And I have found the magical numbers of power-resistors and potentiometers.
